ABSTRACT

Acute hepatotoxicity caused by plants poisoning is responsible for economic losses in farm animals in Brazil. Reports of Cestrum intermedium natural poisoning in cattle are not commonly described in Rio Grande do Sul (RS). This study aimed to document an outbreak of spontaneous C. intermedium poisoning in dairy cattle in the Central-Eastern Mesoregion of RS. Three nine-month-old Holstein and Jersey heifers were affected after they were placed in a small paddock with shortage forage. In this area, specimens of C. intermedium Sendtn with signs of consumption were observed. Morbidity and lethality rates were 100% and clinical courses ranged from 9 to 12 hours. At post mortem examination of the three heifers, there was predominance of acute liver lesions. The liver was moderately enlarged and on the cut surface there was a marked accentuation of the lobular pattern and hemorrhage. Inside the rumen, partially digested C. intermedium Sendtn leaves were observed. The histological aspects of the liver were mostly centrilobular coagulative necrosis and hemorrhage, frequently extended to the midzonal region. The immunohistochemistry technique was performed, in which the polyclonal antibody caspase 3 was used in liver fragments. Moderate to marked immunolabeling was observed in the cytoplasm and nucleus of hepatocytes, predominantly on the periphery of areas of hepatic necrosis indicating cell apoptosis. The diagnosis of C. intermedium Sendtn poisoning in dairy cattle in this study was based on epidemiological, clinical and anatomopathological findings. Since the C. intermedium poisoning is uncommon in dairy cattle, we are describing it for the first time in the Central-Eastern Mesoregion of RS, and represents a differential diagnosis of other acute toxic liver diseases in cattle.(AU)


Intoxicações por plantas que causam hepatotoxicidade aguda são responsáveis por prejuízos econômicos em animais de produção no Brasil. Relatos de intoxicações naturais por Cestrum intermedium em bovinos não são comumente descritos no Rio Grande do Sul (RS). Este trabalho teve como objetivo documentar um surto de intoxicação espontânea por C. intermedium em bovinos leiteiros ocorrido na região Centro-Oriental do RS. Três bovinos com nove meses de idade, das raças Holandês e Jersey, foram acometidos depois de serem introduzidos em um pequeno piquete com escassez de forragem. Nessa área foram observados exemplares de Cestrum intermedium Sendtn com sinais de consumo. As taxas de morbidade e letalidade foram de 100% e os cursos clínicos variaram de 9 a 12 horas. Na necropsia dos três bovinos, observou-se predomínio de lesões hepáticas agudas. O fígado dos animais acometidos estava moderadamente aumentado de tamanho e na superfície de corte havia marcada acentuação do padrão lobular e hemorragia. Na abertura do rúmen, foram observadas folhas de Cestrum intermedium Sendtn parcialmente digeridas. Histologicamente, o fígado apresentava necrose coagulativa centrolobular e hemorragia. Frequentemente, essas alterações se estendiam para a região mediozonal. Foi realizada a técnica de imuno-histoquímica, na qual utilizou-se o anticorpo policlonal caspase 3 em fragmentos de fígado. Notou-se marcação moderada a acentuada em citoplasma e núcleo de hepatócitos, predominantemente na periferia das áreas de necrose hepática indicando apoptose celular. O diagnóstico de intoxicação por Cestrum intermedium Sendtn nos bovinos leiteiros desse estudo, foi realizado com base nos achados epidemiológicos, clínicos e anatomopatológicos. A intoxicação por C. intermedium é incomum em bovinos leiteiros, sendo descrita pela primeira vez na região centro oriental do RS e representa um diagnóstico diferencial de outras hepatopatias tóxicas agudas.(AU)

ABSTRACT

Serum blood samples from 50 Murrah buffalo calves were examined in this study. The animals were allocated into three groups according to the number of parturitions of their mothers: G1 (n= 15) calves from primiparous buffaloes, G2 (n= 19) calves from buffaloes with two to four parturitions, and G3 (n= 16) calves from buffaloes with five or more parturitions. Blood samples were taken at birth, before colostrum ingestion, at 24h, 48h, and 72h after birth, and at 7, 14, 21, and 30 days after birth for determination of levels of gammaglutamyl transferase (GGT), alkaline phosphatase (ALP), aspartate aminotransferase (AST), creatine kinase, total protein, albumin, globulins (including immunoglobulin G), iron, total calcium, ionized calcium, phosphorus, sodium, and potassium. The age of the calves was found to influence all of the biochemical parameters, with the exception of ionized calcium and potassium in the calves in groups G1 and G3. The calving order was found to influence AST, GGT, total protein, albumin, and globulins, including IgG. The high serum ALP activity in the first two days after birth indicates that measurement of the levels of this enzyme may be used as an indirect method of assessing passive immunity transfer.(AU)


Amostras de sangue de 50 bezerros de búfalo Murrah foram examinados nesse estudo. Os animais foram distribuídos em três grupos de acordo com a paridade de suas genitoras: G1 (n=15) bezerros de búfalas primíparas, G2 (n=19) bezerros de búfalas com 2 a 4 gestações, e G3 (n=16) bezerros de búfalas com cinco ou mais gestações. Amostras de sangue foram colhidas ao nascimento, antes da ingestão de colostro e 24h, 48h, e 72h após o nascimento e 7, 14, 21 e 30 dias após nascimento para determinar níveis de gammaglutamil transferase (GGT), fosfatase alcalina (ALP), aspartato aminotrasferase (AST), creatina quinase, proteínas totais, albumina, globulina (inclusive imunoglobulina G), ferro, cálcio total, cálcio ionizado, fósforo, sódio e potássio. A idade dos bezerros influenciou todos os parâmetros bioquímicos, exceto cálcio ionizado e potássio nos bezerros dos grupos G1 e G3. A ordem de nascimento influenciou AST, GGT, proteínas totais, albumina e globulinas, inclusive IgG. Intensa atividade ALP no soro nos primeiros dois dias após nascimento indica que medidas dos níveis dessa enzima podem ser utilizados como método indireto de avaliar transferência passiva de imunidade.(AU)

ABSTRACT

O objetivo deste estudo longitudinal foi avaliar a influência da vacinação materna na transferência de anticorpos (ACs) contra as viroses respiratórias em bezerros. Para tanto, vacas e bezerros foram distribuídos em dois grupos conforme a realização (VAC, n=6) ou não (NVAC, n=4) da vacinação no pré-parto. Amostras sanguíneas foram obtidas após a parição (D0); em seguida, apenas os bezerros foram acompanhados até D180. ACs séricos foram determinados pela vírus-neutralização (VN) contra BVDV, BoHV-1, BRSV e BPI3-V. Vacas VAC apresentaram diferenças para ACs contra BoHV-1 (P=0,01) em D0. As frequências (%) de bezerros soropositivos para BoHV-1 foram maiores em VAC do D2 ao D120; para BRSV em D4, D8, D10 e D150 (P≤0,08); medianas de ACs contra BoHV-1 em VAC do D2 ao D120 (P=0,08). A vacinação das vacas no pré-parto foi fundamental para a transferência e a duração de ACs contra BoHV-1 e BRSV, porém o protocolo adotado não foi eficiente para o aumento de ACs para BVDV e BPI3-V. A eficácia parcial da vacinação materna não inviabiliza a sua recomendação devido à importância do Herpesvírus e do BRSV na DRB, porém estratégias para a melhoria nas respostas imunes contra as demais viroses devem ser estabelecidas.(AU)


The aim of this longitudinal research was to evaluate the influence of maternal vaccination for the passive immune transfer of antibodies (Abs) against respiratory viruses in calves. Therefore, cows and calves sourced from two groups according to having received (VAC, n=6) or not (NVAC, n=4) of vaccination at pre-partum period. Blood samples of cows and calves harvested after parturition (D0), and then only calves followed until the age of 180 days. Abs performed by virus neutralization (VN) against BVDV, BoHV-1, BRSV and BPI3-V. VAC cows presented differences for Abs against BoHV-1 (P=0.01) in D0. Frequencies (%) of seropositive VAC calves for BoHV-1 were higher in D2 to D120; to BRSV D4, D8, D10 and D150 (P≤0.08); higher medians of Abs against BoHV-1 in VAC at D2 up to D120 (P=0.08). Partial efficacy of vaccination of cows does not impair its recommendation due to the importance of Herpesvirus and BRSV in BRD, but strategies for improvement in immune responses against other viruses should be established.(AU)

Esta pesquisa avaliou a TIP e a dinâmica de anticorpos (ACs) específicos em bezerros naturalmente expostos aos agentes causadores da doença respiratória bovina (DRB). Foram selecionados 19 bezerros Holandeses alimentados com colostro proveniente de doadoras vacinadas para DRB. Amostras de soro foram obtidas antes e após a ingestão do colostro (48h) para a soroneutralização (SN). Os valores médios (log2) detectados após colostragem foram de 11,5±1,6 (BVDV), 8,8±1,3 (BoHV-1), 5,5±1,6 (BRSV) e 8,4±1,5 (BPIV-3). Cinco bezerros foram criados do nascimento aos 240 dias de vida, observando-se decréscimo nos títulos de ACs para BVDV, BoHV-1 e BPIV-3 ao longo do tempo (P≤0,001). As taxas de infecções detectadas entre o D14 e o D240 foram de 40% (2/5), 20% (1/5), 80% (4/5), e 60% (3/5), respectivamente, para BVDV, BoHV-1, BRSV e BPIV-3. A maioria dos bezerros manifestou broncopneumonia após as infecções virais. Os bezerros apresentaram ACs para todas as viroses às 48 horas de vida, porém os títulos adquiridos para o BRSV foram baixos. A susceptibilidade para as infecções variou de acordo com os níveis e a duração dos títulos de ACs maternos.(AU)


This research evaluated the PIT and the dynamics of specific antibody (Ab) for calves naturally exposed to the viral agents involved in Bovine Respiratory Disease (BRD). Nineteen Holstein calves fed colostrum from vaccinated donors for DRB. Serum samples were obtained before and after colostrum intake (48h) for serum neutralization (SN). Mean values (log2) detected after colostrum feeding were 11.5±1.6 (BVDV), 8.8 ±1.3 (BoHV-1) 5.5±1.6 (BRSV) and 8.4±1.5 (BPIV-3). Five calves were raised from birth to 240 days of life and presented a decrease in Ab titers for BVDV, BoHV-1 and BPIV-3 over time (P≤ 0.001). Infection rates from D14 to D240 were of 40% (2/5), 20% (1/5), 80% (4/5) and 60% (3/5), respectively for BVDV, BoHV-1, BRSV and BPIV-3. Most of the calves presented bronchopneumonia after seroconversion to the virus. Calves presented Ab for all viruses at 48 hours of life, however BRSV Ab titer were low. Levels and persistence of maternal antibody titers determined the susceptibility to viral infections.(AU)

ABSTRACT

The hematological and immunological development of calves from birth to 6 months of age was performed by hemogram and cellular phenotype. Ten male Holstein calves were assessed in 13 moments: before colostrum intake (D0), every 2 days until the 10th day of life (D2 to D10), at the 15th day after birth (D15), and then monthly up to 6 months (D180). Calves presented hemoconcentration on day (D) 0 and showed a gradual decrease in hematimetric rates until D180. The inversion of the neutrophilic for lymphocytic profile was observed on D4. The percentage of CD3+ cells on D10 was higher than D30 up to D180. The number of CD4+ and CD8+ cells did not change between time points. The number of CD21+ lymphocytes was significantly higher at early time points of D0 up to D15, compared on D30 until D150. In conclusion, the neonatal period was marked by stress leukogram in the first 4 days, and low number of B lymphocytes. These might be risk factors for bacterial infections responsible for navel inflammation and diarrhea. The increase in the number of B cells from 30 days of age demonstrated that the calves were functional and able to generate an immune response.(AU)


O desenvolvimento hematológico e o imunológico de bezerros do nascimento aos seis meses de idade foram avaliados por meio hemograma e imunofenotipagem dos linfócitos. Para tanto, 10 bezerros machos Holandeses foram avaliados em 13 momentos: antes da ingestão do colostro (D0), a cada dois dias até o 10º dia (D2 a D10), 15º dia (D15) e, em seguida, mensalmente até seis meses de idade (D180). Os bezerros apresentaram hemoconcentração em D0 e decréscimo gradual dos índices hematimétricos até D180. A inversão do perfil neutrofílico para linfocítico foi observada em D4. Os valores de CD3+ (%) em D10 foram maiores que aqueles observados em D30 até D180. CD4+ e CD8+ não apresentaram alterações entre os momentos. Os linfócitos B CD21+ aumentaram significativamente a partir dos primeiros momentos D0 até D15, em comparação com os momentos subsequentes D30 até D150. Em conclusão, o período neonatal foi caracterizado pelo leucograma de estresse nos primeiros quatro dias e o baixo número de linfócitos B. Esses fatos podem ser considerados como fatores de risco para infecções bacterianas responsáveis pelas inflamações umbilicais e pela diarreia. Por outro lado, o aumento de células B após os 30 dias de vida demonstrou que os bezerros foram capazes de gerar uma resposta imune.(AU)

This study evaluated the expression of CD14, toll-like receptor (TLR) 2 and TLR4 on the surface of milk neutrophils in bovine mammary glands infected with Corynebacterium bovis. Here, we used 23 culture-negative control quarters with no abnormal secretion on the strip cup test and milk somatic cell count lower than 1x105 cells/mL, and 14 C. bovis infected quarters. The identification of neutrophils, as well as, the percentage of neutrophils that expressed CD14, TLR2 and TLR4 were analyzed by flow cytometry using monoclonal antibodies. The present study encountered no significant difference in the percentages of milk neutrophils that expressed TLR2 and TLR4 or in the expression of TLR4 by milk neutrophils. Conversely, a lower median fluorescence intensity of TLR2 in milk neutrophils was observed in C. bovis-infected quarters. The percentage of neutrophils that expressed CD14 and the median fluorescence intensity of CD14 in milk neutrophils was also lower in C. bovis-infected quarters...


O presente estudo objetivou avaliar alterações na expressão de CD14, e dos receptores do tipo toll (TLR) 2 e 4 na superfície de neutrófilos lácteos provenientes de glândulas mamárias infectadas por Corynebacterium bovis. O presente estudo utilizou 23 quartos negativos no exame bacteriológico, sem alterações na prova de fundo escuro e com contagem automática de células somáticas menor que 1 x105 células/mL, e 14 quartos mamários infectados por C. bovis A identificação de neutrófilos, assim como a porcentagem de neutrófilos lácteos que expressaram CD14, TLR2 e 4 foram avaliadas por citometria de fluxo utilizando anticorpos monoclonais. A porcentagem de neutrófilos que expressaram TLR2 e TLR4 nos quartos mamários infectados por C. bovis não diferiu dos quartos mamários sadios, assim como na expressão de TLR4. No entanto, a intensidade de fluorescência do TLR2 na superfície dos neutrófilos foi menor nos quartos mamários infectados por C. bovis. A porcentagem de neutrófilos que expressaram CD14 e a intensidade de fluorescência da molécula de CD14 foi menor na superfície dos neutrófilos lácteos dos quartos infectados por C. bovis...

ABSTRACT

Ten male, 12-month-old Jersey with intact spleens, serologically and parasitologically free from Babesia were housed individually in an arthropod-free isolation system from birth and throughout entire experiment. The animals were randomly divided into two groups. Five animals (group A) were intravenously inoculated with 6.6 X10(7) red blood cells parasitized with pathogenic sample of Babesia bovis (passage 7 BboUFV-1), for the subsequent "ex vivo" determination of the expression of adhesion molecules. Five non-inoculated animals (group B) were used as the negative control. The expression of the adhesion molecules ICAM-1, VCAM, PECAM-1 E-selectin and thrombospondin (TSP) was measured in bovine umbilical vein endothelial cells (BUVECs). The endothelial cells stimulated with a pool of plasma from animals infected with the BboUFV-1 7th passage sample had a much more intense immunostaining of ICAM-1, VCAM, PECAM-1 E-selectin and TSP, compared to the cells which did not received the stimulus. The results suggest that proinflammatory cytokines released in the acute phase of babesiosis may be involved in the expression of adhesion molecules thereby implicating them in the pathophysiology of babesiosis caused by B. bovis.


Dez bezerros machos, da raça Jersey, com 1 ano de idade com baços "in situ", sorológica e parasitologicamente livres de Babesia, foram mantidos em baias individuais no isolamento a prova de artrópodes do Depto de Veterinária desde o nascimento e ao longo de toda a experimentação. Os animais foram divididos aleatoriamente em dois grupos. Cinco animais (grupo A) foram inoculados por via intravenosa com 6,6 x10(7) hemácias parasitados com amostra patogênica de Babesia bovis (BboUFV - 1 7ª passagem) , para a determinação subseqüente "ex vivo" da expressão de moléculas de adesão . Cinco animais não inoculados (Grupo B ) foram utilizados como controlo negativo . A expressão de moléculas de adesão ICAM - 1, VCAM , PECAM - 1, E - selectina e trombospondina ( TSP ) foi medida em células endoteliais da veia umbilical de bovinos (BUVECs). As células endoteliais estimuladas com um pool de plasma proveniente de animais infectados com BboUFV - 1 7ª passagem tinham uma imunocoloração muito mais intensa de ICAM - 1 , VCAM , PECAM - 1 de E - selectina e de TSP , em comparação com as células que não receberam o estímulo . Os resultados sugerem que as citocinas pró-inflamatórias liberados na fase aguda da babesiose pode estar envolvida na expressão de moléculas de adesão , implicando , assim, elas na fisiopatologia da babesiose causada por B. bovis.

ABSTRACT

The aim of this study was to evaluate the seroprevalence of leptospirosis in the dairy herds from Minas Gerais, Brazil, during the years 2009 and 2010. A total of 2,915 serum samples were collected from the lactating cows of 151 properties in eleven municipalities located in the Sete Lagoas region. The Microscopic Agglutination Test was used to detect antileptospiral agglutinins. An individual animal prevalence of 20.7 percent (95 percent CI = 17.1 percent - 24.3 percent) and a herd prevalence of 80.8 percent (95 percent CI = 73.8 percent = 87.7 percent) were determined. The most prevalent serovars were hardjoprajitno at 19.4 percent; hardjoprajitno strain Norma at 17.4 percent; and hardjo-bovis at 17.4 percent. These results show the significance of the hardjo serovar in bovine leptospirosis cases in Minas Gerais...


Avaliou-se a prevalência de aglutininas antileptospira em rebanhos leiteiros durante os anos de 2009 e 2010. Foram coletadas 2.915 amostras de soro de vacas lactantes de 151 propriedades localizadas na região de Sete Lagoas - MG. Foi utilizado o teste de aglutinação microscópica (MAT) para detecção de aglutininas antileptospira. Prevalência animal de 20,7 por cento (95 por cento CI = 17,1 por cento-24,3 por cento) e prevalência de rebanho de 80,8 por cento (95 por cento CI = 73,8 por cento = 87,7 por cento) foram reportadas. Os sorotipos mais prevalentes no estudo foram hardjoprajitno (19,4 por cento); hardjoprajitno estirpe Norma (17,4 por cento) e hardjo-bovis (17,4 por cento). Estes resultados demostram a importância da sorovariedade hardjo nos casos de leptospirose bovina do estado de Minas Gerais...

ABSTRACT

Este trabalho foi desenvolvido com o objetivo de avaliar a resistência aos ectoparasitas em bovinos jovens da raça Crioula Lageana e meio-sangue Angus em condições naturais. Foram utilizados 10 machos castrados em cada grupo, recém desmamados (6-8 meses) e mantidos sob as mesmas condições de manejo, em pastagens cultivadas de inverno e verão, no município de Monte Castelo - SC, sofrendo infestação natural por ectoparasitas. A cada 28 dias, fêmeas de carrapatos (Rhipicephalus (Boophilus) microplus) com tamanho acima de 4 mm foram quantificadas nos dois lados do corpo de cada animal, bem como os nódulos com larvas de Dermatobia hominis. A espessura da capa do pelame foi também avaliada a cada coleta e os animais foram classificados quanto à coloração do pelame. Os animais da raça Crioula Lageana foram mais resistentes às infestações por D. hominis e R. microplus. Não houve diferença entre a disposição de carrapatos e bernes conforme os lados direito e esquerdo dos animais. Os animais de pelagem escura albergaram a maior porcentagem de ectoparasitas nos grupos avaliados.


This study was carried out to evaluate the breed resistance against ectoparasites infestations in Crioula Lageana and crossbred Angus male calves under natural condition. Ten weaned and neutered male calves (6-8 months) of each group were kept together in winter and summer pastures in Monte Castelo, Santa Catarina State. Every 28 days, female ticks (Rhipicephalus (Boophilus) microplus) more than 4 mm long were counted on the right and left side of the body of each animal, as well as the D. hominis nodules. Coat thickness also was measured at each sampling and all animals were classified according coat color. Crioula Lageana group was less infested by D. hominis than crossbred Angus in five times and by ticks in the infestation peak (P<0.05). There were no differences in the distribution of the ticks and grubs between animal body sides. Dark-haired animal in both groups were the most susceptible.

ABSTRACT

A total of 184 serum samples were collected including; 92 from slaughtered cattle [23/ season] in Abo El Matameer Abattoir, and 92 from random human patients attending a private laboratory in Abo El Matameer District, Behera Province to be examined for presence of antibodies against F. hepatica. Cattle samples were examined by Distomiasis Fumouze kits[registered] [ready to use indirect haemagglutination test kits] and it was found that the total prevalence was 9.8%. The seasonal prevalence of F. hepatica infection in examined serum samples of cattle revealed that winter and spring seasons showed the highest seasonal prevalence followed by summer season and males [10.8%] were higher than females [7.4%]. In addition, the highest prevalence was observed in the age group 2 - 4 years [18.18%] followed by the age group > 4 years [5.88%]. On the other side, the overall detection rate of F. hepatica infection in human was 13.04% and the highest seasonal prevalence was noticed in winter season [21.74%] followed by summer and spring season. The sexual distribution of the positive reactors to F. hepatica in human revealed higher prevalence in females [13.3%] than in males [12.9%] and the age group 20 - < 40 years [24.4%] showed the highest prevalence followed by the age group 40 - < 60 years [22.7%]

ABSTRACT

Rapid diagnostic tests are needed to facilitate diagnosis and control of brucellosis. Sero-epidemiology of brucellosis is currently done by employing the Rose Bengal test [RBT]. Also the world organization for animal health [OIE] has approved an indirect ELISA for testing serum and milk. The Immuno-Chromatographic brucellosis test [ICT] is a rapid, card-based immunochromatographic test for detection of antibodies directed against B. abortus antigens. To the best of our knowledge, ICT has not been used for the diagnosis of brucellosis in cattle yet. The objective of the study was to evaluate the performance of the ICT brucellosis test for the diagnosis of B. abortus in cattle sera versus RBT and ELISA as a gold standard, Also evaluation of ICT brucellosis test efficacy for detection of antibodies against B. Abortus in milk samples. 94, 90.36 and 84.3% of sera samples were positive by RBT, ICT and ELISA respectively. The ICT had 94.44% sensitivity and 100% specificity versus RBT and ELISA as a gold standard. The kappa value between ICT and ELISA was 0.76. Also ICT detected antibodies against B. abortus in serum and milk samples of 7 cows while a recently calved cow showed antibodies in the milk sample only. In conclusion, ICT provides a handy and accurate tool for the rapid detection of antibodies to B. abortus in cattle sera and milk

ABSTRACT

Despite our current knowledge of the immunology, pathology, and genetics of Anaplasma marginale, prevention in cattle is currently based on old standbys, including live attenuated vaccines, antibiotic treatment, and maintaining enzootic stability in cattle herds. In the present study, we evaluated the use of an immunostimulant complex (ISCOMATRIX) adjuvant, associated with a pool of recombinant major surface proteins (rMSP1a, rMSP1b, rMSP4 and rMSP5) to improve the humoral immune response triggered in calves mainly by IgG2. Ten calves were divided in three groups: 4 calves were inoculated with the ISCOMATRIX/rMSPs (G1); 2 calves were inoculated with ISCOMATRIX adjuvant (G2); and 4 calves received saline (G3). Three inoculations were administered at 21-day intervals. In G1, the calves showed significant increases in total IgG, IgG1 and IgG2 levels 21 days after the second inoculation, compared to the control group (p < 0.05), and G1 calves remained above the cut-off value 28 days after the third inoculation (p < 0.05). The post-immunized sera from calves in G1 reacted specifically for each of the rMSPs used. In conclusion, the ISCOMATRIX/rMSPs induced antigen-specific seroconversion in calves. Therefore, additional testing to explore the protection induced by rMSPs, both alone and in conjunction with proteins previously identified as subdominant epitopes, is warranted.


Apesar dos avanços da imunologia, patologia e genética de Anaplasma marginale, a prevenção em bovinos ainda é baseada nas vacinas vivas atenuadas, na terapia com antibiótico e estabilidade enzoótica dos rebanhos bovinos. No presente estudo, avaliou-se o uso de um complexo imunoestimulante (ISCOMATRIX), associado às proteínas recombinantes de superfície (rMSP1a, rMSP1b, rMSP4 e rMSP5) para melhorar a resposta imune humoral desencadeada em bezerros, principalmente por IgG2. Dez animais foram divididos em três grupos: 4 bezerros foram inoculados com o ISCOMATRIX/rMSPs (G1), 2 bezerros foram inoculados com ISCOMATRIX adjuvante (G2) e 4 bezerros receberam salina (G3). Três doses vacinais foram administradas em intervalos de 21 dias. No G1, os bezerros apresentaram aumentos significativos nos níveis de IgG total, IgG1 e IgG2 21 dias após a segunda inoculação, em comparação com o grupo de controle (p <0,05). Nos bezerros do G1 esses níveis de anticorpos permaneceram acima do ponto de corte 28 dias após a terceira inoculação (p < 0,05). Os soros pós-imunização de bezerros do G1 reagiram especificamente com cada uma das rMSPs utilizadas. Em conclusão, o ISCOMATRIX/rMSPs induziu soroconversão antígeno-específica em bezerros. Portanto, se justifica a realização de ensaios adicionais para explorar a proteção induzida pela rMSPs, tanto sozinhas como em conjunto com novas proteínas identificadas com epítopos subdominantes.

ABSTRACT

The plants which cause sudden death of cattle in Brazil occupy a leading position for losses in the cattle industry. Amorimia exotropica is one of the plants pertaining to this group. Diagnostic findings in these cases may be inconclusive; further knowledge is necessary. This paper identifies cardiac lesions through anti-cardiac troponin C (cTnC) immunehistochemistry performed in tissues from cattle poisoned after consumption of A.exotropica in southern Brazil. Heart fragments from nine A. exotropica-poisoned cattle were studied immunohistochemically using anti-human cTnC as the primary antibody. In the hearts from all of the poisoned cattle, there was a sharp decrease in the cTnC expression level in the cytoplasm of groups of cardiomyocytes. A significant decrease in anti-cTnC immunoreactivity occurred particularly in degenerated or necrotic cardiomyocytes. Occasional groups of cells showed complete loss of immunolabeling. In the remaining intact cardiomyocytes from poisoned cattle and in cardiomyocytes from six cattle that died from other causes there was intense cytoplasmic staining.


No Brasil, plantas cujo consumo determina morte súbita de bovinos estão entre as principais causas de perdas na pecuária. Esse trabalho identifica lesões cardíacas através de imuno-histoquímica antitroponina cardíaca C (TncC), desenvolvida em tecidos de bovinos intoxicados após consumo de Amorimia exotropica, no sul do Brasil. Fragmentos cardíacos de nove bovinos intoxicados, naturalmente, por Amorimia exotropica foram examinados por imuno-histoquímica anti-TncC, como anticorpo primário. Nos corações de todos os bovinos intoxicados pela planta, havia pronunciada diminuição dos níveis de expressão de TncC no citoplasma de grupos de cardiomiócitos. Diminuição significativa na imunorreatividade anti-TncC ocorreu, particularmente, em cardiomiócitos degenerados ou necróticos. Grupos ocasionais de células mostraram completa perda de imunomarcação. Em cardiomiócitos remanescentes e intactos de bovinos intoxicados e em cardiomiócitos de seis bovinos que morreram por outras causas, observou-se intensa coloração citoplasmática.

The objective of this study was to investigate the prevalence of anti-Neospora caninum antibodies in cattle from milk producing farms of the microregion of Batalha, state of Alagoas, Brazil, as well as to identify the risk factors associated with the infection. Blood samples were collected from 1,004 cattle of 17 farms for the serological investigation regarding the presence of anti-N. caninum antibodies by the Indirect Immunofluorescence Reaction Technique (IMRT). From the total amount of samples analyzed, 77/1,004 (7.67%) were positive and 927/1,004 (92.33%) were negative. The logistical regression identified that cattle from farms without consortium breeding have an infection risk 6.33 (p<0.001; C.I. 2.89-13.10) times higher than cattle from farms with that type of breeding. Cattle from farms where the aborted fetuses are not adequately buried have an infection risk 3.04 (p<0.001; C.I. 1.64-5.63) times higher than cattle from farms with adequate destination of these fetuses. Infection by N. caninum occurs in cattle of the investigated region. The factors identified in our study can be used as risk indicators, so that control measures could be implemented to avoid infection by N. caninum in the herds of this region.


Objetivou-se com este estudo investigar pos anti-N. caninum através do teste de Reação de Imunoflua prevalência de anticorpos anti-Neospora caninum em bovi-orescência Indireta (RIFI). Do total das amostras analisadas, nos procedentes de propriedades leiteiras da microrregião 77/1004 (7,67%) foram positivas e 927/1004 (92,33%) fo-Batalha, Estado de Alagoas, Brasil, além de identificar os fato-ram negativas. A regressão logística identificou que animais res de risco associados à infecção. Foram coletadas amostras de sangue de 1004 bovinos procedentes de 17 propriedades para investigação sorológica quanto à presença de anticorpos anti-N. caninum através do teste de Reação de Imunofluorescência Indireta (RIFI). Do total das amostras analisadas, 77/1004 (7,67%) foram positivas e 927/1004 (92,33%) foram negativas. A regressão logística identificou que animais de propriedades sem criação consorciada têm risco 6,33 (p<0,001; I.C. 2,89-13,10) vezes maior de infecção do que animais de propriedades onde ocorre esse tipo de criação. Animais de propriedades onde os fetos abortados não são adequadamente enterrados têm risco 3,04 (p<0,001; I.C. 1,64- 5,63) vezes maior de infecção do que animais de propriedades onde é feito o destino adequado dos mesmos. A infecção por N. caninum ocorre em bovinos na região estudada. Os fatores identificados neste estudo podem servir como indicadores de risco para que sejam implantadas medidas de controle para evitar a infecção por N. caninum nos rebanhos dessa região.

ABSTRACT

Descreve-se um surto de intoxicação por Pterodon emarginatus em bovinos no Estado de Goiás. De um lote de 84 bovinos que comeram avidamente folhas e os frutos da planta após a queda acidental de uma árvore, todos os animais adoeceram e sete morreram. Os sinais clínicos observados foram eriçamento dos pelos, retração do flanco, apatia profunda, prostração, tremores musculares, ressecamento do focinho, tenesmo, incoordenação, relutância em movimentar-se e decúbito esternal prolongado. As atividades séricas de AST, ALT e GGT e os teores de bilirrubina estavam acentuadamente elevados. As principais alterações macroscópicas consistiam de hepatomegalia e áreas multifocais de necrose no fígado, além de hemorragias no coração, pleura parietal, mesentério, omento, serosa do rúmen, baço, pulmão, subcutâneo e musculatura esquelética intercostal e torácica. Microscopicamente, observou-se degeneração e necrose hepatocelular massiva moderada a acentuada, hiperplasia biliar multifocal moderada e bilestase multifocal leve. Adicionalmente, notou-se degeneração vacuolar multifocal moderada nos túbulos contorcidos dos rins.


An outbreak of poisoning by Pterodon emarginatus in cattle in Goiás, Brazil, is described. Eighty four cattle that accidentally consumed the leaves and fruits from a P. emarginatus tree, fallen in a storm, were affected and seven died. Clinical signs included piloerection, flank retraction, marked apathy, prostration, muscle tremors, muzzle dryness, tenesmus, incoordination, reluctance to move, and prolonged sternal recumbency. AST, ALT, and GGT serum activity, and bilirubin levels were markedly increased. Gross lesions included hepatomegaly, multifocal areas of necrosis in the liver, and hemorrhages in the heart, parietal pleura, mesentery, omentum, ruminal serosa, spleen, lung, subcutaneous tissue, and thoracic and intercostal skeletal muscles. Histologically, moderate to severe, massive hepatocellular degeneration and necrosis with moderate bile duct hyperplasia and mild bile stasis were observed. In addition, moderate multifocal vacuolar degeneration in the convoluted renal tubular cells was noted.

Passive immunity transfer (PIT) evaluation is an essential tool for the maintenance of healthy calves during the first months of life. Since lactation number and breed have been proven to influence immunoglobulin levels in colostrum, the aim of this study was to evaluate PIT from primiparous and multiparous Canchim cows to their calves. Blood samples were collected from the calves before colostrum intake and 1, 2, 7, 15 and 30 days thereafter, while colostrum samples from the cows were taken immediately after parturition. Activities of gamma-glutamyl transferase (GGT), alkaline phosphatase (ALP), and concentrations of total protein, albumin, globulins, immunoglobulin A (IgA), immunoglobulin G (IgG), total and ionized calcium, inorganic phosphorus, magnesium, sodium and potassium were evaluated in calves' serum and activities of GGT and ALP and concentrations of total protein, IgA and IgG were assessed in cow's colostrum whey. Immunoglobulins concentrations were evaluated by electrophoresis in polyacrylamide gels. Serum biochemistry evaluations revealed an increase in gamma-glutamyl transferase and alkaline phosphatase activities and in total protein, globulins, immunoglobulin A and immunoglobulin G levels in calves' serum after colostrum intake. Only total protein and light chain immunoglobulin G levels in colostrum whey were affected by the cows' lactation number. Phosphorus and magnesium levels in blood serum increased after colostrum intake, while sodium and potassium levels oscillated in the experimental period. PIT was influenced by the cows' lactation number but was efficient in both groups.


A avaliação da transferência de imunidade passiva (TIP) é uma ferramenta essencial para manutenção de bezerros saudáveis nos primeiros meses de vida. Uma vez que há influência do número de lactações e da raça de vacas nos teores de imunoglobulinas do colostro, o presente estudo foi conduzido com o objetivo de avaliar a TIP de vacas Canchim primíparas e pluríparas aos seus bezerros. Amostras de sangue dos bezerros foram coletadas antes da ingestão de colostro e 1, 2, 7, 15 e 30 dias após o nascimento e amostras de colostro das vacas foram coletadas imediatamente após o parto. As atividades de gamaglutamiltransferase (GGT), fosfatase alcalina (ALP) e as concentrações de proteína total, albumina, globulinas, imunoglobulina A (IgA) e imunoglobulina G (IgG), cálcio total e ionizado, fósforo, magnésio, sódio e potássio foram avaliadas no soro dos bezerros e as atividades de GGT e ALP e as concentrações de proteína total, IgA e IgG foram avaliadas no soro colostral. A concentração de imunoglobulinas foi avaliada por meio de eletroforese em gel de poliacrilamida. As avaliações bioquímicas do soro sanguíneo dos bezerros revelaram aumento nas atividades das enzimas gamaglutamiltransferase e fosfatase alcalina e nos teores de proteína total, globulinas, imunoglobulina A e imunoglobulina G após a ingestão do colostro. Apenas os teores de proteína total e imunoglobulina G de cadeia leve no soro colostral foram influenciados pelo número de lactações das vacas. Os teores de fósforo e magnésio aumentaram após a ingestão de colostro, enquanto as concentrações de sódio e potássio oscilaram no decorrer do período experimental. A TIP foi influenciada pelo número de lactações das vacas, no entanto mostrou-se eficiente em ambos os grupos.

ABSTRACT

Conglutinin is a high molecular-weight lectin originally detected in bovine serum. It belongs to the family of collectins that bind sugar residues in a Ca(2+)-dependent manner and are effector molecules in innate immunity. Conglutinin appears to play an important role in immune defense mechanisms, showing antiviral and antibacterial activities when tested in vivo and in vitro. The present study evaluated the effect of conglutinin on the respiratory bursts in bovine peripheral phagocytes. Using nitroblue tetrazolium and hydrogen peroxide assays, we showed that sugar ligand-bound conglutinin stimulated the production of superoxide and H2O2 in granulocytes whereas the non-sugar-bound form of conglutinin inhibited these processes. These results indicate that both forms of conglutinin are able to interact with surface leukocyte receptors but have opposite effects on phagocytic activity. Our findings suggest that conglutinin bound to sugar residues on microbial surfaces can induce oxygen burst in phagocytes, and thereby mediates the elimination of pathogens and prevents the spread of infection.

ABSTRACT

The purpose of our study was to investigate changes in immunological parameters induced by weaning stress (including milk restriction) in calves. Fifteen Holstein calves were subjected to weaning at 6 weeks of age. Blood samples were collected at -14, -7, -2, 1, 3, and 5 days post-weaning (DPW; 0 DPW = 42 days). Weaning caused significant (p < 0.01) increases in the neutrophil (NE):lymphocyte (LY) ratio at 5 DPW with a significant (p < 0.05) reduction of LYs. The concentration of acute-phase proteins (haptoglobin and serum amyloid A) also increased significantly (p < 0.05) at 3 and 5 DPW compared to -2 DPW. Levels of the iron-binding protein lactoferrin decreased significantly (p < 0.05) after weaning. Serum tumor necrosis factor-alpha and cortisol levels were elevated (p < 0.05) at 3 DPW, while those of serum interferon-gamma decreased (p < 0.05) at 1 and 3 DPW compared to levels observed before weaning. Weaning significantly (p < 0.05) decreased the percentage of CD25+ T cells in the peripheral blood. In conclusion, weaning stress affected the NE:LY ratio along with the levels of acute phase proteins, lactoferrin, cortisol, and inflammatory cytokines in the peripheral blood of calves. Weaning stress may induce an acute phase response possibly through the elevation of cortisol production and modulation of inflammatory cytokines.

The serological response of beef calves was evaluated with different vaccination regimens against blackleg, using an official strain (MT) and a field-collected strain of Clostridium chauvoei as antigens. Sixty calves were randomly allocated to four different groups and were submitted to distinct vaccination protocols with a commercial polyvalent vaccine. Group G1 was first vaccinated at four months of age and a booster shot was given after weaning, at eight months. Group G2 was given the first dose at eight months and a booster shot 30 days later. Group G3 was vaccinated only once at eight months and the control group was not vaccinated. These alternative vaccination regimens were proposed in an effort to adequately protect cattle under open-field farming conditions. Serological evaluations were made by Elisa at 4, 8, 9 and 10 months of age. Both groups receiving booster shots had a significantly increased serological response 30 days later. However, the serum IgG levels against C. chauvoei were significantly higher in the calves that were first vaccinated at four months. At 10 months, the two booster shot groups (G1 and G2) had similar serological responses, while the calves that were treated with a single dose of vaccine at weaning (G3) had a response that was similar to that of the control group. The serological response of the calves was significantly inferior at several of the evaluation times when the field strain of the bacteria was used as a challenge antigen instead of the official MT strain. The serological response of calves that are vaccinated twice was found to be satisfactory, independent of the first injection being made at four or eight months of age. It was also concluded that it would be useful to include local bacterial strains in commercial vaccine production.


Foi avaliada a resposta sorológica de bezerros de corte submetidos a diferentes esquemas de vacinação contra o carbúnculo sintomático, empregando-se como antígenos duas cepas distintas de Clostridium chauvoei: uma oficial (MT) e a outra uma cepa de campo. Os animais (n=60) foram randomizados em quatro grupos (G1, G2, G3 e Controle) e submetidos a três protocolos distintos de vacinação com um produto comercial polivalente. O G1 foi primovacinado aos 4 meses de idade e recebeu o reforço após desmama (8 meses de idade). O G2 recebeu a primeira dose aos 8 meses de idade e reforço 30 dias após. O G3 foi vacinado somente aos 8 meses de idade e o Controle não foi vacinado. As avaliações sorológicas pelo ensaio imunoenzimático (Elisa) foram realizadas aos 4, 8, 9 e 10 meses de idade dos bezerros. Nos dois esquemas em que os animais receberam o booster (G1 e G2), houve um aumento significativo (p<0,05) da resposta sorológica quando foram avaliados 30 dias após. No entanto, os valores séricos de IgG contra C. chauvoei foram significativamente maiores nos animais primovacinados aos 4 meses de idade. Aos 10 meses, os dois grupos que receberam o reforço vacinal (G1 e G2) não diferiram entre si na resposta sorológica e os bezerros que receberam uma única dose de vacina na desmama (G3) não diferiu do Controle. A resposta sorológica dos bezerros foi significativamente inferior (p<0,05) em diversos momentos da avaliação, quando a cepa de campo foi empregada como antígeno e quando comparada à da cepa MT. Pode-se deduzir que a resposta sorológica dos bezerros vacinados aos 4 e 8 meses de idade foi satisfatória e que existiram diferenças significativas nos valores séricos de anticorpos contra C. chauvoei quando na avaliação foi empregada cepa de campo.

ABSTRACT

Apesar de ser alto o número de acidentes ofídicos em bovinos, as alterações hematológicas e bioquímicas séricas sãopouco estudadas, especialmente nas três primeiras semanas após o envenenamento. Diante deste contexto, objetivou-senesta pesquisa caracterizar o perfil hematológico e bioquímico sérico de bovinos após a inoculação experimental do venenode Bothrops alternatus. Cinco novilhas mestiças de peso corporal entre 188 e 278kg e idade de 12 a 18 meses receberamna altura média da face cranial do membro anterior direito, entre as articulações umerorradioulnar e do carpo, por viaintramuscular superficial, 0,15mg/kg de veneno de Bothrops alternatus diluído em salina. Coletou-se sangue de todos osanimais antes (tempo zero) e as 3, 8, 24, 48, 72, 96, 168, 240, 405 e 576 horas após inoculação do veneno para avaliaçãodo hemograma, coagulograma e dosagem de proteína total, albumina, globulinas, ureia, creatinina, creatina fosfotransferase,aspartato aminotransferase e gama glutamiltransferase. O veneno de B. alternatus causou intoxicação grave nos bovinos,e foi letal para dois animais às 53 horas e 78 horas. As alterações hematológicas observadas foram: anemia normocíticanormocrômica, leucocitose com neutrofilia com desvio para esquerda regenerativo, linfocitose, aumentos dos tempos decoagulação, de protrombina e de tromboplastina parcial ativada, trombocitopenia e elevação da ureia e das enzimas CPT eAST.


Despite the high number of Bothrops envenomation in cattle, the hematological and biochemical changes are poorly known mainly in the first few weeks after the poisoning. In this context, the aim of this study was to evaluate the bovine blood profile after B. alternatus envenomation. Five crossbred heifers were inoculated with B. alternatus venom (0.15mg/kg) diluted in saline solution, by intramuscular injection, in the mean height of the cranial side of right forelimb, between the joints umerradio-ulnar and carpus. The blood was collected before the beginning of the experiment (zero time) and at 3, 8, 24, 48, 72, 96, 168, 240, 405 e 576 hours after inoculation, in order to evaluate the hemogram, coagulogram, total protein, albumine, globulins, urea, creatinine, creatinine phosphokinase (CPT) and aspartate aminotransferase. The B. alternatus venon caused a serious envenomation in catlle and was lethal for two animals at 53 and 78 hours. The blood profile alteratons were normocitic and normocromic anaemia, leucocitosis with neutrophilia, linfocitosis, coagulation time increased, trombocytopenia and elevation in the urea, CPT and AST enzymes.
